Histamine and 5-Hydroxytryptamine on Isolated Atrial Preparation of Tortoise

Abstract
1. In isolated atrial preparation of tortoise, effects of histamine and 5-hydroxytryptamine(5-H T) were investigated.
2. The responses to histamine and 5-HT showed seasonal variation. In general the atria from summer tortoise responded with augmentation to both agents, the atria from winter tortoise with inhibition.
3. The inhibitory response to histamine was not related to parasympathetic mechanism, but abolished or markedly reduced by minute dose of an antihistaminic drug(phenergan). This suggests existence of specific histamine receptor which responds with inhibition to histamine.
4. The inhibitory response to 5-HT. was-antagonized by atropine and hexamethonium, indicating that the inhibition is originated from stimulation of the parasympathetic ganglia in the atria.
5. The excitatory response to histamine was not related to adrenergic mechanism. The response was inhibited by an antihistammic(pbenergan), suggesting existence of specific histamine receptor which responds with augmentation.
6. The excitatory respose to 5-HT was also not related to adrenergic mechanism. An antiserotonin(methysergide) did not abolish the 5-HT effect but reduced it.
